目的:建立以高效液相色谱法测定人血清中万古霉素浓度的方法。方法:血清样品以20%偏磷酸溶液沉淀蛋白,离心后取上清液直接进样测定。其中,色谱柱为Hypersil C18,流动相为乙腈-0.0125mol·L-1磷酸二氢钾缓冲液(10∶90),流速为0.8mL·min-1,柱温为25℃,检测波长为236nm。结果:万古霉素血药浓度在2.0~170.0mg·L-1范围内线性关系良好(r=0.9998),最低检测浓度为2.0mg·L-1;低、中、高(2.5、30.8、150.4mg·L-1)3种浓度的平均相对回收率分别为99.8%、101.1%、98.9%,日内及日间RSD均<2.5%(n=5)。结论:本方法简便、准确、快速,适用于人血清万古霉素浓度的监测和药动学研究。
Objective: To establish a method for the determination of vancomycin in human serum by high performance liquid chromatography. Methods: Serum samples were precipitated with 20% meta-phosphoric acid solution, centrifuged and the supernatant was directly injected into the sample. The column was Hypersil C18, the mobile phase was acetonitrile-0.0125 mol·L-1 potassium dihydrogen phosphate buffer (10:90), the flow rate was 0.8 mL · min-1, the column temperature was 25 ℃ and the detection wavelength was 236 nm . Results: The linearity of vancomycin in the range of 2.0-170.0 mg · L-1 was good (r = 0.9998) and the lowest concentration was 2.0 mg · L-1. The low, middle and high (2.5, 30.8, 150.4 mg · L-1) were 99.8%, 101.1% and 98.9%, respectively. The intra-day and inter-day RSD were all less than 2.5% (n = 5). Conclusion: The method is simple, accurate and rapid and suitable for the monitoring and pharmacokinetic study of human serum vancomycin.
