将体外培养良好的NIT-1细胞分为对照组、油酸组、软脂酸组、油酸+软脂酸组,37℃、5%CO2条件下培养72h,放免法测定培养液上清中胰岛素含量,RT-PCR法检测细胞胆囊收缩素受体CCK-AR、CCK-BR mRNA的表达。结果:经FFAs处理的NIT-1细胞培养液胰岛素含量明显降低,油酸组更显著;油酸、软脂酸、油酸+软脂酸处理后细胞的CCK-AR mRNA表达明显低于对照组。结论:游离脂肪酸可显著减少小鼠胰岛β细胞CCKAR mRNA的表达以及降低胰岛素的分泌量。
NIT-1 cells cultured in vitro were divided into control group, oleic acid group, palmitic acid group, oleic acid + palmitate group, cultured in 37 ℃, 5% CO2 for 72h, radioimmunoassay Insulin content, CCK-AR and CCK-BR mRNA expression were detected by RT-PCR. Results: The content of insulin in NIT-1 cells treated with FFAs was significantly lower than that in oleic acid groups. The expressions of CCK-AR mRNA in oleic acid, palmitic acid and oleic acid + palmitate were significantly lower than those in control group . Conclusion: Free fatty acids can significantly reduce the expression of CCKAR mRNA in pancreatic β-cells and decrease insulin secretion.
