目的观察不同浓度的三氧化二砷作用不同时间对小儿恶性畸胎瘤细胞的影响,分析其作用机制。方法体外原代培养小儿恶性畸胎瘤细胞,应用不同浓度的三氧化二砷作用后,用噻唑蓝〔3-(4,5-dimethyl-thia zol-2-y1)-2,5-diphenyl tetrazolium bromide MTT〕比色法观察其对恶性畸胎瘤细胞增殖的影响,应用脱氧核糖核苷酸末端转移酶介导的缺口末端标记法(TUNEL)及流式细胞仪检测三氧化二砷对恶性畸胎瘤细胞凋亡的诱导作用。结果不同浓度的三氧化二砷作用于小儿恶性畸胎瘤细胞有明显的时间和剂量依赖性,三氧化二砷作用后细胞生长明显受抑制,并出现明显的凋亡特征性改变,经TUNEL法可检测到细胞凋亡现象,经流式细胞仪可检测到小儿恶性畸胎瘤细胞在G1峰前出现明显的凋亡峰。结论三氧化二砷可明显抑制小儿恶性畸胎瘤细胞的生长,其机制主要可能是诱导恶性畸胎瘤细胞凋亡。
Objective To observe the effect of different concentrations of arsenic trioxide on malignant teratoma cells in children at different times and to analyze its mechanism. Conclusion Arsenic trioxide can significantly inhibit the growth of malignant teratoma cells in children, and its mechanism may mainly induce the apoptosis of malignant teratoma cells.
