目的:研究不同剂量内皮素受体A拮抗剂FR139317对大鼠心力衰竭模型的作用。方法:结扎雄性Wistar大鼠左冠状动脉致心肌梗死并充血性心力衰竭,给予FR1393171和5mg·kg~(-1)·d~(-1)两种剂量治疗6周,观察内皮素受体A拮抗剂对心衰大鼠血液动力学、血浆内皮素水平及死亡率的作用,并比较不同剂量间的差异。结果:二种不同剂量FR139317治疗均改善心衰大鼠的血液动力学,降低血浆内皮素水平,并降低死亡率。较高剂量FR139317明显降低心衰大鼠的平均动脉压,并使血浆内皮素的浓度降至更接近于心功能正常大鼠的水平。结论:选择性内皮素受体A拮抗剂FR139317可有效地治疗心力衰竭。
Objective: To study the effect of different doses of endothelin receptor A antagonist FR139317 on heart failure model in rats. Methods: Left ventricular myocardial infarction and congestive heart failure were induced in male Wistar rats. Two doses of FR1393171 and 5 mg · kg -1 · d -1 were administered for 6 weeks. Endothelin A Antagonists on hemodynamics, plasma endothelin levels and mortality in heart failure rats, and compare the differences between different doses. Results: Two different doses of FR139317 treatment ameliorate hemodynamics, decrease plasma endothelin level and reduce mortality in heart failure rats. Higher doses of FR139317 significantly reduced mean arterial pressure in heart failure rats and decreased plasma endothelin concentrations closer to normal heart function rats. Conclusion: The selective endothelin receptor A antagonist FR139317 can effectively treat heart failure.
