采用国产JQ-700型基因枪,以含Bt基因的质粒pHB101为供体,轰击小麦、玉米愈伤组织,轰击后的愈伤组织经潮霉素抗性筛选后,获得32株小麦再生苗,14株玉米抗性植株。以a-32P-dCTP标记的PHB101质粒为探针,对小麦抗性植株进行点杂交印渍检测,发现有15株小苗显示阳性信号,且其中有一株的后代对田间蚜虫及锈病的抗性较好。玉米再生植株种植田间后在其后代中选出一抗玉米螟的稳定株系,尚需进一步的分子验证和饲虫实验。
Using domestic JQ-700 gene gun, Bt gene containing plasmid pHB101 as donor, bombarding wheat, corn callus, after bombardment callus by hygromycin resistance screening, obtained 32 wheat seedlings, 14 maize-resistant plants. A-32P-dCTP-tagged PHB101 plasmid was used as a probe to detect dot blot hybridization of wheat-resistant plants. Fifteen seedlings showed positive signals, and one of the offspring showed greater resistance to field aphids and rust it is good. After the corn regenerated plants were planted in the field, the stable strains of the resistant corn borer were selected among their offspring, and further molecular verification and feeding experiments were needed.
