目的探讨反义寡核苷酸对胆囊癌细胞端粒酶活性的影响及对胆囊癌细胞生长增殖的作用。方法设计针对端粒酶RNA亚基模板序列并经硫代磷酸修饰的反义、正义和随机序列寡核苷酸,并以正常人成纤维细胞作对照,观察其对人胆囊癌细胞(GBCSD)端粒酶活性和细胞生长增殖的影响作用以及对正常人成纤维细胞的作用。结果反义寡核苷酸可有效地抑制胆囊癌细胞的端粒酶活性,呈剂量依赖性,并明显的抑制胆囊癌细胞的生长,对正常人成纤维细胞生长无明显作用。结论硫代反义寡核苷酸(PSODN)对胆囊癌端粒酶活性有明显的抑制用并促进细胞凋亡。
Objective To investigate the effect of antisense oligonucleotide on telomerase activity in gallbladder carcinoma cells and its effect on the proliferation and proliferation of gallbladder carcinoma cells. Methods Antisense, sense and random oligonucleotides modified by phosphorothioate were designed according to the template sequence of telomerase RNA subunit. The normal human fibroblasts were used as controls to observe the effect on human gallbladder carcinoma cells (GBCSD) The role of telomerase activity and cell growth and proliferation and normal human fibroblasts. Results Antisense oligodeoxynucleotides could effectively inhibit the telomerase activity of gallbladder carcinoma cells in a dose - dependent manner and significantly inhibit the growth of gallbladder carcinoma cells and had no effect on the growth of normal human fibroblasts. Conclusion Sulfo antisense oligonucleotide (PSODN) can inhibit the telomerase activity of gallbladder carcinomas and promote apoptosis.
