胱硫醚β- 合酶催化同型半胱氨酸与丝氨酸缩合成胱硫醚,后者经茚三酮显色后在465 nm 波长处有最大吸收峰,而底物丝氨酸和同型半胱氨酸与茚三酮的反应产物在此波长吸收很低,因此可用酶促反应后465 nm 吸光度的增加值检测胱硫醚β- 合酶活性的高低。依据这一原理,本文建立了胱硫醚β- 合酶活性比色测定方法。实验结果发现,该方法具有精密度高、重复性好和简便易行等优点。对大鼠不同组织胱硫醚β- 合酶活性进行测定的结果显示,大鼠各种组织中均有胱硫醚β-合酶分布,但其活性存在一定差异。
Cystathionine β-synthase catalyzes the condensation of homocysteine with serine to form cystathionine, the latter having the maximum absorption peak at 465 nm after being developed by ninhydrin, whereas the substrate serine and homocysteine The reaction product with ninhydrin has a very low absorption at this wavelength, so the level of cystathionine β-synthase activity can be measured using an increase in absorbance at 465 nm after enzymatic reaction. Based on this principle, this paper established a cystathionine β- synthase activity colorimetric assay. Experimental results show that the method has the advantages of high precision, good repeatability and easy to operate. The assay of cystathionine β-synthase activity in different tissues of rats showed that cystathionine β-synthase was distributed in various tissues of rats, but its activity was different.
