目的:研究雷公藤红素对HER2/neu过表达人类乳腺癌MDA-MB-453细胞周期的影响。方法:流式PI单染检测细胞周期的分布;Western Blot检测细胞周期相关蛋白的改变。结果:用3.00、6.00、12.00μM雷公藤红素处理MDA-MB-453细胞24 h后,细胞周期阻滞在G0/G1期,并检测到该周期相关蛋白Cyclin D1、P27、p-Rb均发生了变化,且呈浓度依赖性改变。结论:雷公藤红素诱导MDA-MB-453细胞周期停止于G0/G1期,且与P27上调,Cyclin D1蛋白水平下调及Rb的高磷酸化状态下降有关。
Objective: To study the effects of tripterine on the cell cycle of HER2 / neu overexpressing human breast cancer cell line MDA-MB-453. Methods: The distribution of cell cycle was detected by flow cytometry. The changes of cell cycle related proteins were detected by Western Blot. Results: The cell cycle arrest was in G0 / G1 phase after being treated with 3.00, 6.00, 12.00 μM tripterine for 24 h, and the expression of Cyclin D1, P27 and p-Rb Has changed, and the concentration-dependent changes. Conclusions: Tripterine induced the cell cycle arrest of G0 / G1 phase in MDA-MB-453 cells, and was related to the up-regulation of P27, the down-regulation of Cyclin D1 protein and the decrease of Rb hyperphosphorylation.
