采用MTT比色分析法和流式细胞仪检测不同浓度的雌三醇,不同浓度的他莫昔芬以及雌三醇加他莫昔芬对存在雌激素受体的HT-29人结肠癌细胞株增殖的影响,结果显示雌三醇能提高细胞增殖率,其浓度对数值与此增殖率呈显著正相关(r=0.975,P<0.005),细胞周期各时相分布显示S期细胞数目明显增加;他莫昔芬有抑制细胞增殖作用,其浓度值与此增殖率呈显著负相关(r=-0.957,P<0.05);他莫昔芬对雌三醇的增殖作用有部分抑制。结果提示雌三醇对雌激素受体阳性的HT-29细胞株有增殖效应,这种效应能被雌激素受体拮抗剂他莫昔芬部分阻断,为临床用他莫昔芬治疗雌激素受体阳性的结肠癌提供了依据。
Different concentrations of estriol, tamoxifen at different concentrations, and estriol plus tamoxifen were tested for HT-29 human colon cancer cells in the presence of estrogen receptor using MTT colorimetric assay and flow cytometry.
