目的 研究丁酸钠体外诱导人脑多型性胶质母细胞瘤细胞系 BT-325细胞的分化。方法 以 1mmol/L丁酸钠处理 BT-325细胞 ,测定细胞生长曲线 ,流式细胞术分析细胞周期, Western blot分析 BT-325中 GFAP的表达变化 ,鉴定细胞的分化状态。结果 BT-325细胞经丁酸钠诱导 6~ 8 d后,出现明显的分化特征:胞体显著增大,贴壁明显,细胞伸出多个突起,与相邻的细胞接触,且有接触抑制现象;细胞生长显著受抑 ,80%的细胞被阻断在 G1和 S期;胶质纤维酸性蛋白的含量明显增加。结论 经 1mmol/L丁酸钠处理后 ,人脑多型性胶质母细胞瘤细胞系 BT-325细胞发生一定程度的分化。
Objective To investigate the differentiation of BT-325 cells induced by sodium butyrate in human brain glioblastoma multiforme cell line in vitro. Methods BT-325 cells were treated with 1 mmol / L sodium butyrate and the cell growth curve was determined. The cell cycle was analyzed by flow cytometry. The expression of GFAP in BT-325 was analyzed by Western blot, and the cell differentiation status was identified. Results BT-325 cells showed obvious differentiation after induced by sodium butyrate for 6-8 days: the cell bodies were significantly increased, the adherent cells were obvious, the cells extended a number of protrusions and contacted with adjacent cells, and the phenomenon of contact inhibition ; Cell growth was significantly inhibited, 80% of the cells were blocked in the G1 and S phase; glial fibrillary acidic protein content increased significantly. Conclusion BT-325 cells of human brain glioblastoma multiforme cell line differentiated to a certain extent after being treated with 1 mmol / L sodium butyrate.
