目的 研究胰岛素对大鼠全脑缺血再灌注损伤的治疗时间窗 .方法 造成 42只大鼠全脑缺血后 ,于再灌注后 0 ,30 min,1,3,6 ,9和 12 h等不同时间点脑室内注射胰岛素 1U,再灌注 7d后取脑进行病理检查 ,比较不同处理对大鼠海马 CA1区细胞的影响 .结果 再灌注 0 min时给予胰岛素的保护作用最为强烈 ,随着给予胰岛素时间的推迟 ,胰岛素对海马 CA1区细胞的保护作用逐渐减弱 .再灌注 6 h给予胰岛素海马 CA1区正常神经元计数为 (86± 8) mm- 1 ,是正常对照组的 48.0 % .再灌注 9h后给予胰岛素 ,海马 CA1区神经元大部分已坏死 ,与缺血对照组无显著性差异 (P>0 .0 5 ) .结论 胰岛素治疗大鼠全脑缺血再灌注损伤的治疗时间窗为 6 h
Objective To study the therapeutic time window of insulin on global cerebral ischemia-reperfusion injury in rats.Methods 42 rats were subjected to 0, 30 min, 1, 3, 6, 9 and 12 h after reperfusion The intracerebroventricular injection of 1U of insulin at different time points and the 7-day reperfusion followed by brain pathological examination were performed to compare the effects of different treatments on the hippocampal CA1 region of rats.Results The protective effect of insulin on 0 min reperfusion was the strongest, The delayed protection of insulin in hippocampal CA1 area gradually decreased, and the number of normal neurons in hippocampal CA1 area at 6 h after reperfusion was (86 ± 8) mm- 1, which was 48.0% of that in normal control group. After reperfusion for 9h Most of the neurons in the hippocampal CA1 area were necrotic, and there was no significant difference with the ischemic control group (P> 0.05) .Conclusion The therapeutic window of global ischemia-reperfusion injury in insulin-treated rats is 6 h
