在人体衰老过程中,机体各系统不断地发生变化,其中免疫系统的变化较为明显。淋巴细胞是免疫系统的主要细胞,研究其功能变化对研究衰老及抗衰老都具有重要意义。为此,我们选用外周血淋巴细胞为实验对象,用紫外线照射,造成细胞DNA损伤,在加入羟基脲抑制DNA半保留复制合成的前提下,通过~3H-TdR参入率测定人淋巴细胞DNA期外合成(un-scheduled DNA synthesis,UDS)的变化,以此来观察和比较不同年龄的人淋巴细胞DNA对损伤的修复能力。 1材料与方法 1.1细胞老年人无肿瘤和内分泌病病史,青年人为献血者,取出的静脉血肝素抗凝,然后用淋巴细胞分离液(上海试剂二厂)分离出淋巴细胞。将细胞加到含植物血凝素(PHA)的1640培养液中,培养3天后测定
