目的:采用病、证模型复合的方法建立大鼠肾亏型颈椎病动物模型。方法:选择3月龄雌性SPF级SD大鼠30只,随机分为正常组、颈椎病模型组和肾虚型颈椎病模型组,每组10只。采用动静力失衡性大鼠颈椎间盘退变模型复合去卵巢肾虚亏模型的方法建立肾虚型颈椎病模型。通过检测大鼠子宫及附件形态和质量、血清雌二醇(estradiol,E2)含量验证肾虚证;通过颈椎间盘组织病理学、Ⅱ型和Ⅹ型胶原的免疫组化检测及聚集蛋白聚糖(aggrecan-1,Agc1)、Ⅱ型前胶原基因(typeⅡprocolla-gen gene,Col2a1)、基质金属蛋白酶-13(matrix metalloproteinase-13,MMP-13)和基质金属蛋白酶抑制剂-1(tissue inhibitor of metalloproteinases-1,TI MP-1)的基因表达情况评判椎间盘退变。结果:与正常组和颈椎病模型组相比,肾虚型颈椎病模型组动物子宫及附件形态萎缩、质量减轻,血清雌二醇含量降低,颈椎间盘组织病理学退变更加明显,Ⅱ型胶原蛋白表达减少,Ⅹ型胶原表达增高,Agc1和Col2a1基因表达降低,MMP-13基因表达增高。结论:通过病、证模型复合的方式建立了肾虚型颈椎病模型,肾虚可以加重颈椎间盘的退变。
OBJECTIVE: To establish an animal model of renal deficiency type cervical spondylosis by using compound disease and syndrome model. Methods: Thirty female Sprague-Dawley rats of 3 months old were randomly divided into normal group, cervical spondylosis model group and kidney deficiency type cervical spondylosis model group with 10 in each group. The animal model of cervical syndrome with deficiency of kidney was established by the method of combining dynamic and static imbalance of rat model of cervical disc degeneration and ovariectomized kidney deficiency model. The morphology and quality of rat uterus and appendages were detected, and the serum levels of estradiol (E2) were assayed for renal deficiency syndrome. Immunohistochemistry and immunohistochemical staining of aggrecan (type Ⅱ and type Ⅹ) -1, Agc1), typeⅡprocolla-gen gene (Col2a1), matrix metalloproteinase-13 (MMP-13) and tissue inhibitor of metalloproteinase-1 , TI MP-1) gene expression assessment of disc degeneration. Results: Compared with the normal group and the cervical spondylosis model group, the uterus and appendix of the kidney deficiency type cervical spondylosis model were atrophied, the quality was reduced, the serum estradiol level was decreased, the histopathological degeneration of cervical intervertebral disc was more obvious, the type Ⅱ collagen Decreased expression of type X collagen, Agc1 and Col2a1 gene expression decreased, MMP-13 gene expression increased. Conclusion: The model of kidney deficiency type of cervical spondylosis was established through the combination of disease and syndrome model. Kidney deficiency can aggravate the degeneration of cervical disc.
