目的:探讨维拉帕米(Ver)增加癌细胞内光敏剂和光动力学疗法(PDT)杀伤作用的效果。方法:在结肠癌细胞培养的基础上,分别采用荧光分光光度法和噻唑蓝(MTT)法分析癌细胞内光敏剂含量和PDT后细胞存活率。结果:Ver可显著增加光敏剂进入癌细胞内的含量,给药后16小时最明显。单独Ver(0.02μg/ml)对癌细胞存活率无明显影响。激光照射后8,16小时,PSD+Ver组细胞存活率与PSD组相比显著降低,32小时细胞存活率接近;加光敏剂后16小时,激光照射,癌细胞存活率较低,可达满意效果。结论:Ver可显著增加癌细胞内光敏剂含量和光动力学杀伤结肠癌细胞能力。
Objective: To investigate the effect of verapamil (Ver) on increasing the photosensitizer and photodynamic therapy (PDT) killing in cancer cells. Methods: Based on colon cancer cell culture, fluorescence spectrophotometry and thiazolyl blue (MTT) method were used to analyze the content of photosensitizer in cancer cells and cell survival rate after PDT. RESULTS: Ver significantly increased the content of the photosensitizer in the cancer cells, which was most pronounced at 16 hours after administration. Ver (0.02 μg/ml) alone had no significant effect on the survival rate of cancer cells. After 8 and 16 hours of laser irradiation, the survival rate of PSD+Ver group was significantly lower than that of PSD group, and the survival rate of 32-hour cells was similar. After 16 hours plus photosensitizer, the survival rate of cancer cells was lower with satisfactory results. Conclusion: Ver can significantly increase the content of photosensitizer in cancer cells and photodynamic killing ability of colon cancer cells.
