目的:研究抗白汤对实验性白癜风豚鼠的治疗作用。方法:采用5%氢醌制备实验性白癜风豚鼠模型,以白癜风胶囊(0.24 g·kg-1)作对照,分别用低、中、高剂量(3.1,6.2,12.4 g·kg-1)抗白汤灌胃,每天1次,连续50 d,相当于造模时间5倍治疗。肉眼观察各药物组治疗实验性白癜风豚鼠模型的疗效,并观察表皮基底细胞和棘细胞黑色素分布以及对豚鼠模型血液中胆碱酯酶(CHE)、单胺氧化酶(MAO)、丙二醛(MDA)含量的影响;各药物组对豚鼠模型黑色素吸光度及酪氨酸酶含量和血液流变学各项指标的影响。结果:造模后,模型组血浆黏度、全血黏度,CHE,MAO,MDA含量明显升高,毛囊中黑色素数量、吸光度以及酪氨酸酶含量明显减少,与正常组比较均有显著性差异(P<0.01)。抗白汤各剂量组与模型组比较,能显著增加皮肤表皮基底细胞和棘细胞黑色素分布和皮肤毛囊中黑色素(P<0.01),能显著增强黑色素的吸光度以及增加酪氨酸酶的含量(P<0.01),能显著降低血液中CHE,MAO,MDA含量(P<0.01),能显著改善血浆黏度及全血黏度(P<0.01),并呈现良好的量-效关系,与白癜风胶囊组比较也有显著性差异(P<0.05)。结论:抗白汤对实验性白癜风豚鼠模型具有良好的治疗作用,其主要机制可能与促进血液循环,降低血液中CHE,MAO,MDA含量和增加酪氨酸酶含量,增加毛囊中黑色素数量,增加其吸光度等有关。
Objective: To study the anti-Bai Tang on experimental vitiligo treatment of guinea pigs. Methods: Experimental vitiligo guinea pig model was prepared by using 5% hydroquinone. The vitiligo capsules (0.24 g · kg -1) were used as controls. The low, medium and high doses (3.1, 6.2, 12.4 g · kg -1) Gavage once a day for 50 days, equivalent to 5 times the modeling time treatment. The therapeutic effect of each drug group on guinea pigs with experimental vitiligo was observed with the naked eye. The distribution of melanin in basal cells and spine cells in epidermis and the content of CHE, MAO and MDA in guinea pig model were observed. The effect of each drug group on the melanin absorbance and tyrosinase content of guinea pig model and various indicators of hemorheology. Results: After model establishment, the plasma viscosity, whole blood viscosity, the content of CHE, MAO and MDA in the model group were significantly increased, the amount of melanin, the absorbance and the tyrosinase content in hair follicle were significantly decreased, which were significantly different from those in normal group P <0.01). Compared with the model group, the anti - Bai Tang decoction could significantly increase the melanin distribution in the epidermal basal cells and spine cells and the melanin in the skin follicles (P <0.01), and could significantly increase the absorbance of melanin and increase the content of tyrosinase (P < 0.01). It could significantly reduce the contents of CHE, MAO and MDA in blood (P <0.01), significantly improve the plasma viscosity and whole blood viscosity (P <0.01), and showed a good dose-effect relationship with Vitiligo capsule group Significant difference (P <0.05). CONCLUSION: Anti-Bai Decoction has a good therapeutic effect on guinea pig model of experimental vitiligo. Its main mechanism may be to promote blood circulation, reduce CHE, MAO, MDA content in the blood and increase the content of tyrosinase, increase the number of melanin in hair follicles, Absorbance and so on.
