目的和方法:观察猪肝部分切除后24、48和72h颈外静脉及72h肝静脉和门静脉的血清,以及术后72h从再生肝制得的肝细胞生长因子(pigregeneratinghepatocytegrowthfactor,PRHGF)和它们的不同浓度对体外原代培养肝细胞生长的影响。结果:加入术后上述静脉血清均具有明显的促进体外培养肝细胞生长的作用。其中肝静脉血清的作用最强、门静脉血清的次之,颈外静脉血清的作用居第三,而且当加入浓度上升至80%时,它们的促肝细胞生长作用与50%浓度相比均无显著降低,相反,加入术前颈外静脉血清浓度达80%时,其刺激肝细胞生长作用则明显降低。PRHGF的促进肝细胞生长作用在一定范围内呈剂量依赖性增强。当加入浓度为034mg蛋白质/孔时,达到顶峰,此后即逐渐下降。加入浓度为049mg蛋白质/孔时,则出现抑制效应。结论:术前正常猪颈外静脉血清和PRHGF除有刺激肝细胞生长作用外,尚有抑制肝细胞生长的效应,当所用浓度增加达到一定值时,即对原代培养肝细胞生长呈现抑制效应
