目的:建立测定红细胞胆固醇含量的方法。方法:采用高效液相色谱法,色谱柱为Boston Green ODS,C18(4.6 mm×250 mm,5μm)120 A。流动相为乙腈-异丙醇(85∶15),流速为1.0mL·min-1,检测波长为210 nm。结果:胆固醇在19.69~393.8μg·mL-1范围内呈良好的线性关系(Y=4 418.4 X-39 172,r=0.999 8),精密度、稳定性、重复性均良好,RSD低于2.0%,平均加样回收率为98.8%,RSD为2.47%(n=9)。结论:该方法简便、快速、准确、专属性强、重复性好,可用于红细胞中胆固醇含量测定。
Objective: To establish a method for the determination of red blood cell cholesterol content. Methods: High Performance Liquid Chromatography (HPLC) was used. The column was Boston Green ODS, C18 (4.6 mm × 250 mm, 5 μm). The mobile phase was acetonitrile-isopropanol (85:15), the flow rate was 1.0mL · min-1, the detection wavelength was 210 nm. Results: Cholesterol showed good linearity (Y = 4 418.4 X-39 172, r = 0.999 8) in the range of 19.69-393.8 μg · mL-1, with good precision, stability and reproducibility. RSD was lower than 2.0 %, The average recovery was 98.8%, RSD was 2.47% (n = 9). Conclusion: The method is simple, rapid, accurate, specific and reproducible. It can be used for the determination of cholesterol in erythrocytes.
