目的了解宁夏部分地区蚊虫种类组成和不同蚊种细胞色素氧化酶亚基Ⅰ(COI)基因特征和蚊虫分子系统进化关系。方法对宁夏银川市、石嘴山市、吴忠市和青铜峡市的三带喙库蚊、里海伊蚊、淡色库蚊和八代按蚊4种蚊种COI基因序列进行扩增、测序,在Gen Bank中对所测序列进行相似性比对,分析基因特征、颠换率、遗传距离,利用Mega 7.0软件构建分子系统树。结果本研究中相似性比对结果,其他蚊虫分别与其同种蚊虫的相似性均达到94%~99%,COI基因序列扩增长度为417 bp左右,GC含量为29.4%~31.5%,种间颠换率为5.80%~13.81%,种间遗传距离为0.121~0.160。结论 COI基因具有种间特异性,可作为不同蚊种分类鉴别的参考依据。
Objective To understand the species composition of mosquitoes and the relationship between cytochrome oxidase subunit Ⅰ (COI) gene and molecular phylogenetic relationships of mosquitoes in some areas of Ningxia. Methods The COI gene sequences of four species of Culex tritaeniorhynchus, Aedes albopictus, Culex pipiens pallens and Anopheles sinensis in Yinchuan, Shizuishan, Wuzhong and Qingtongxia of Ningxia were amplified and sequenced. In Gen Bank The similarity of the tested sequences was analyzed, the gene characteristics, transversion rate and genetic distance were analyzed. The molecular phylogenetic tree was constructed by Mega 7.0 software. Results The results showed that the similarities between other mosquitoes and their mosquitoes were 94% -99%, respectively. The length of COI gene was about 417 bp and the GC content was 29.4% -31.5% The transversion rate was 5.80% ~ 13.81%, the interspecific genetic distance was 0.121 ~ 0.160. Conclusion The COI gene has interspecific specificity and can be used as a reference for classification and identification of different mosquito species.
