目的观察血管紧张素Ⅱ(AngⅡ)对正常血压和高血压患者的肠系膜动脉血管平滑肌细胞(VSMC)增殖及细胞内NADPH氧化酶-活性氧信号传导通路的影响。方法选取开腹手术正常血压和高血压患者肠系膜动脉培养的二至四代人VSMC作为标本,以四甲基偶氮唑盐(MTT)法、流式细胞仪、逆转录聚合酶链式反应(RT-PCR)分别测定细胞增殖率、细胞内活性氧及p22phox蛋白mRNA的表达。结果①与空白对照组比较,孵育组的细胞增殖率、细胞内活性氧物质生成量、细胞内p22phoxmRNA的表达均明显增高(均P<0.01)。②与正常血压组比较,给予AngⅡ刺激后,高血压患者的肠系膜动脉VSMC增殖率和细胞内p22phoxmRNA的表达量均明显增高(均P<0.01)。结论AngⅡ对高血压患者肠系膜动脉VSMC促细胞增殖作用较正常血压者更强。
Objective To investigate the effects of angiotensin Ⅱ (Ang Ⅱ) on the proliferation of mesenteric artery smooth muscle cells (VSMCs) and the intracellular NADPH oxidase - reactive oxygen species (ROS) signal transduction pathway in normal and hypertensive patients. Methods VSMCs from two to four generations cultured in mesenteric artery of normotensive and hypertensive patients undergoing open surgery were selected as samples and analyzed by MTT assay, flow cytometry and reverse transcriptase polymerase chain reaction RT-PCR) were used to determine the cell proliferation rate, intracellular reactive oxygen species and p22phox mRNA expression. Results ① Compared with the blank control group, the cell proliferation rate, intracellular reactive oxygen species production and intracellular p22phox mRNA expression were significantly increased (all P <0.01). ②Compared with normotensive group, the proliferation rate of mesenteric artery VSMC and the expression of p22phox mRNA in hypertensive patients were significantly increased after AngⅡ stimulation (all P <0.01). Conclusion Ang Ⅱ is more effective than normotensive in proliferating mesenteric artery VSMC in hypertensive patients.
