目的观察环氧化酶2(COX2)抑制剂塞来昔布对戊四氮诱发大鼠癫痫持续状态(SE)后脑内血管活性肠肽(VIP)表达的影响。方法用免疫组化法测定戊四氮诱发癫痫大鼠模型脑组织海马区VIP的含量,探讨塞来昔布对癫痫脑组织VIP含量的影响及剂量关系。结果模型组海马区VIP含量较正常对照组及给药组明显增多(P<0.05)。随着给药浓度的增加,VIP的含量逐渐降低,各组大鼠脑组织VIP的含量比较差异有统计学意义(P>0.05)。结论SE后脑内VIP含量增加,塞来昔布能够有效减少SE大鼠脑内VIP的表达,其可能通过影响大鼠脑内VIP的含量而减轻癫痫发作,起到脑保护作用。
Objective To investigate the effect of cyclooxygenase 2 (COX2) inhibitor celecoxib on the expression of vasoactive intestinal peptide (VIP) in brain of pentylenetetrazole-induced status epilepticus (SE) rats. Methods Immunohistochemical method was used to determine the content of VIP in the hippocampus of pentylenetetrazole-induced epilepsy rat model and to explore the effect of celecoxib on the VIP content of epileptic brain tissue and the dose relationship. Results The content of VIP in hippocampus of model group was significantly higher than that of normal control group and administration group (P <0.05). With the increase of drug concentration, the content of VIP gradually decreased. The content of VIP in each group of rats was significantly different (P> 0.05). Conclusion The content of VIP in brain increased after SE. Celecoxib could effectively reduce the expression of VIP in the brain of SE rats. It may reduce the seizures and the brain protection by affecting the contents of VIP in the brain of rats.
