目的:建立高效液相色谱法测定血浆中巴洛沙星浓度。方法:采用Hypersil C18色谱柱(200mm×4.6mm,5μm);流动相为0.02mol·L-1磷酸二氢钾(含0.5%三乙胺,pH4.0)-甲醇(62∶38);流速1.0mL·min-1;柱温35℃;检测波长295nm。结果:血浆巴洛沙星在0.05~4.0mg·L-1范围内线性关系良好(r=0.9995)。低、中、高3种浓度相对回收率分别为85.84%,86.46%,89.33%,日内、日间RSD均小于10%。结论:本法可用于巴洛沙星血浆中浓度的测定以及药动学研究。
Objective: To establish a method for the determination of balofloxacin in plasma by high performance liquid chromatography. Methods: Hypersil C18 column (200mm × 4.6mm, 5μm) was used. The mobile phase consisted of 0.02mol·L -1 potassium dihydrogen phosphate (0.5% triethylamine, pH 4.0) -methanol (62:38) 1.0mL · min-1; column temperature 35 ℃; detection wavelength 295nm. Results: There was a good linear relationship between plasma barofloxacin and 0.05 ~ 4.0 mg · L-1 (r = 0.9995). The relative recoveries of low, middle and high concentrations were 85.84%, 86.46% and 89.33%, respectively, and the RSDs were less than 10% in both days and days. Conclusion: This method can be used for the determination of plasma concentration of balofloxacin and pharmacokinetic studies.
