目的:建立以高效液相色谱法测定风湿Ⅱ号合剂中绿原酸含量的方法。方法:色谱柱为Diamonsil C18(200mm×4.6mm,5μm),流动相为乙腈-0.4%磷酸溶液(10∶90),检测波长为327nm。结果:绿原酸的检测浓度在24.2~193.6μg·mL-1范围内与峰面积积分值呈良好的线性关系(r=0.9999);平均回收率为99.23%,RSD=1.05%(n=9)。结论:本法准确、灵敏、重复性好,可用于风湿Ⅱ号合剂的质量控制。
Objective: To establish a method for the determination of chlorogenic acid in rheumatism Ⅱ by HPLC. Methods: The column was Diamonsil C18 (200mm × 4.6mm, 5μm). The mobile phase was acetonitrile-0.4% phosphoric acid solution (10:90). The detection wavelength was 327nm. Results: The concentration of chlorogenic acid in the range of 24.2 ~ 193.6μg · mL-1 showed a good linear relationship with the peak area integral (r = 0.9999). The average recovery was 99.23%, RSD was 1.05% (n = 9 ). Conclusion: This method is accurate, sensitive and reproducible. It can be used for quality control of rheumatism Ⅱ.
