常规食用菌一级菌种培养,均采用PDA培养基。我们尝试用成熟桑椹作培养基原料培养食用菌菌种,获得满意结果。 1 培养基制作 5月桑椹成熟季节采集呈黑紫色桑椹。配方:新鲜桑椹200g,琼脂18~20g,水1000mL。将桑椹加水煮沸5分钟,用双层纱布过滤挤压,滤液中加入(?)后再加热,待琼脂熔化后定容,分装灭菌。 2 接种培养 试验用菌种为平菇、金针菇、蘑菇、香菇四类共9种,接种至桑椹培养基中在25℃下培养。 3 培养结果 所有平菇和蘑菇培养1周菌丝长满培养基表面,尤其是平菇杂17气生菌丝相当旺盛。香菇、白金针菇培养10天后亦长满培养基表面,比用PDA生长迅速。在仅用桑椹单一营养基质时,以上几种食用菌均表现出生长旺势。因此,我们认为桑椹是一种良好的食用菌培养基。在试验中,接种的45管斜面中无一污染。之后又接种10管平菇杂17,在未加棉塞的情况
