目的鉴定新疆皮肤利什曼病病原体。方法用利什曼原虫属特异引物R222和R333,从皮肤利什曼病患者皮肤病变组织、婴儿利什曼原虫和热带利什曼原虫基因组DNA中扩增出一特异SSUrDNA多变区片段,将其克隆到pGEM○R-TEasyVector上,并筛选和鉴定出重组质粒。结果以EcoRⅠ酶切、PCR扩增和Southern杂交证实获得的重组质粒包含SSUrDNA多变区片段。结论皮肤利什曼病病原体、婴儿利什曼原虫和热带利什曼原虫SSUrDNA多变区片段重组质粒的获得,为进一步序列分析比较提供实验材料。
Objective To identify the pathogen of cutaneous leishmaniasis in Xinjiang. Methods A specific SSU rDNA variable region fragment was amplified from the skin lesions of cutaneous leishmaniasis, Leishmania infantis and Leishmania leishmanii genomic DNA using Leishmania specific primers R222 and R333. It was cloned into pGEM® R-TEasyVector and the recombinant plasmid was screened and identified. Results The EcoR Ⅰ digestion, PCR amplification and Southern hybridization confirmed that the recombinant plasmid obtained contained the SSUrDNA variable region fragment. CONCLUSIONS The acquisition of the recombinant plasmid of the skin leishmaniasis pathogen, Leishmania infantum and Leishmania leishmanii SSU rDNA variable region fragment provided the experimental materials for further sequence analysis and comparison.
