目的 :探讨 HSP70在人肝癌细胞中的表达。方法 :不同人肝癌细胞经 42℃休克 2 h,分别于休克后 4h、8h、12 h、16 h、48h收集细胞爬片 ;采用 ENVISION免疫组化法检测 HSP70在细胞中的表达 ;采用间接免疫荧光法定量分析 SMMC- 772 1细胞休克前及休克后 16 h HSP70的表达。结果 :免疫组化和免疫荧光检测显示热休克前后不同人肝癌细胞中 HSP70表达明显增加 ,以 16 h表达达高峰。结论 :不同人肝癌细胞休克前后 HSP70表达的动态研究 ,为今后进行 HSP70抗原肽的提取和纯化以及抗癌免疫研究奠定了基础
Objective: To investigate the expression of HSP70 in human hepatoma cells. METHODS: Human hepatoma cells were shocked at 42°C for 2 h, and cells were collected at 4 h, 8 h, 12 h, 16 h, and 48 h after shock. The expression of HSP70 in cells was detected by ENVISION immunohistochemistry; indirect immunity was used. Fluorescence quantitative analysis of HSP70 expression in SMMC-772 1 cells before shock and 16 h after shock. RESULTS: Immunohistochemistry and immunofluorescence showed that HSP70 expression was significantly increased in different human hepatoma cells before and after heat shock and peaked at 16 h. Conclusion : The dynamic study on the expression of HSP70 before and after shock of different human hepatocellular carcinoma cells lays a foundation for the future extraction and purification of HSP70 antigen peptide and the research of anti-cancer immunity.
