目的:观察扶正解毒颗粒(FJG)对硫酸镍(NiSO4)暴露肾损伤大鼠的保护作用。方法:50只Wistar大鼠采用NiSO42.5 mg.kg-1.d-1ip(连续7 d,此后间日1次)制备肾损伤模型,随机分为NiSO4组(模型组)、FJG高、中、低剂量组(20,10,5 g.kg-1.d-1)、二巯基丁二酸(DMSA,50 mg.kg-1.d-1)组。另设NS组(空白对照)及FJG对照组(FJG 10 g.kg-1.d-1)。各组大鼠ig 1次/d,共4周。分别采用放射免疫法(RIA)、光电比色法及酶法检测尿α1-微球蛋白(α1-MG)、N-乙酰-β-D氨基葡萄糖苷酶(NAG)及尿肌酐(UCr)的变化。结果:NiSO4组尿α1-MG及NAG活性(NAG/UCr)较NS组显著增高(P<0.05和P<0.01),与NiSO4组相比,FJG各组尿α1-MG及NAG活性则明显降低,尤以FJG高、中剂量组为著(P<0.05和P<0.01)。结论:尿α1-MG及NAG活性增高与早期镍性肾损伤存在关联,中药FJG对镍性肾损伤大鼠具有较好的保护作用。
Objective: To observe the protective effect of Fuzheng Jiedu Granule (FJG) on renal injury induced by nickel sulfate (NiSO4) in rats. Methods: Fifty Wistar rats were randomly divided into NiSO4 group (model group), high FJG group (middle model group) and middle model group (model group) with NiSO42.5 mg.kg-1.d-1ip for 7 days, , Low dose group (20,10,5 g.kg-1.d-1) and dimercaptosuccinic acid (DMSA, 50 mg.kg-1.d-1) group. Another NS group (blank control) and FJG control group (FJG 10 g.kg-1.d-1). Each rat ig 1 / d, a total of 4 weeks. The urinary α1-microglobulin (α1-MG), N-acetyl-β-D-glucosaminidase (NAG) and urinary creatinine (UCr) were measured by radioimmunoassay (RIA) Variety. Results: Urinary α1-MG and NAG activity (NAG / UCr) in NiSO4 group were significantly higher than those in NS group (P <0.05 and P <0.01). Compared with NiSO4 group, urinary α1-MG and NAG activity were significantly decreased , Especially in FJG high and medium dose groups (P <0.05 and P <0.01). CONCLUSION: Increased urinary α1-MG and NAG levels are associated with early renal damage in nickel. FJG has a good protective effect on rats with nickel-induced renal injury.
