目的建立同时测定猴耳环药材中杨梅苷(1)、5,3’,4’,5’-四羟基黄烷-7-没食子酸酯(2)、槲皮苷(3)含量的方法,并对不同批次猴耳环药材中3种黄酮的含量进行测定。方法采用Hypersil BDS C18色谱柱(150 mm×4.0 mm,5μm);流动相:0.2%磷酸-乙腈,梯度洗脱;检测波长:320 nm。结果 3种黄酮分离度良好;各成分质量浓度与峰面积在测定范围内均呈良好的线性关系(r≥0.9994);平均加样回收率在99.91%~100.98%之间,RSD为0.36%~1.20%(n=6);所测11批猴耳环药材中3种黄酮(1~3)含量分别在0.36~2.23 mg·g-1(1),6.46~22.41 mg·g-1(2),1.06~4.59 mg·g-1(3)之间。结论该方法简单、准确,具有良好的重复性和稳定性,可作为猴耳环药材的质量控制方法。
Objective To establish a method for simultaneous determination of myricetin (1), 5,3 ’, 4’, 5’- tetrahydroxyflavan-7-gallate (2) and quercitrin (3) The content of three flavonoids in the different parts of the monkey earring herbs was determined. Methods Hypersil BDS C18 column (150 mm × 4.0 mm, 5 μm) was used. The mobile phase was eluted with 0.2% phosphoric acid - acetonitrile. The detection wavelength was 320 nm. Results The separation of three flavonoids was good. The linear relationship was obtained between the mass concentration and the peak area of the three flavonoids (r≥0.9994). The average recovery was between 99.91% and 100.98%, and the RSD was 0.36% 1.20% (n = 6). The contents of three flavonoids (1 ~ 3) in 11 batches of earrings were 0.36-2.23 mg · g-1 (1) and 6.46-22.41 mg · g- , 1.06 ~ 4.59 mg · g-1 (3). Conclusion The method is simple and accurate, with good repeatability and stability, and can be used as a quality control method for monkey earrings.
