目的:探讨砒霜(三氧化二砷ATO)对MRL/lpr自发性狼疮小鼠外周血单个核细胞(PBMC)中DNA甲基转移酶1(DNMT1)和黏附分子淋巴细胞功能相关抗原-1(LFA-1)的亚基CD 11a表达的影响及其可能作用机制。方法:20周龄(发病期)MRL/lpr狼疮小鼠24只,设C57BL正常小鼠24只做参照,随机平分两组,即治疗组(ATO组)和对照组(NS组),治疗组ATO 0.4mg.kg-1.d-1隔日腹腔注射,对照组隔日腹腔注射同等量生理盐水,疗程为60天,采用RT-PCR检测两组小鼠PBMC的DNMT1及CD11a转录水平的表达情况。结果:MRL/lpr小鼠治疗组中DNMT1-mRNA的灰度比值对照组增高(P<0.05),MRL/lpr小鼠治疗组中CD11a-mRNA的灰度比值较对照组降低(P<0.05)。结论:砒霜能逆转系统性红斑狼疮(SLE)的低甲基化状态。
Objective: To investigate the effects of arsenic trioxide (ATO) on DNA methyltransferase 1 (DNMT1) and adhesion molecule lymphocyte-associated antigen-1 (LFA-1) in peripheral blood mononuclear cells (PBMCs) of MRL / Of the subunit CD 11a expression and its possible mechanism of action. Methods: Totally 24 MRL / lpr lupus mice aged 20 weeks (onset stage) were randomly divided into two groups (ATO group and NS group) and 24 C57BL mice (NS group) ATO 0.4mg.kg-1.d-1 was intraperitoneally injected every other day, and the control group was intraperitoneally injected with the same amount of saline on the other day for 60 days. The transcription level of DNMT1 and CD11a in the PBMC of both groups were detected by RT-PCR. Results: The gray value of DNMT1-mRNA in MRL / lpr group was higher than that in control group (P <0.05), and the gray value of CD11a mRNA in MRL / lpr group was lower than that in control group (P <0.05) . Conclusion: Arsenic Sulfate can reverse the hypomethylation status of systemic lupus erythematosus (SLE).
