以甜菜MADS-box基因cDNA片段为探针,采用Southern杂交证实了其在甜菜基因组中为低拷贝序列;利用荧光原位杂交技术(FISH)将其初步定位在二倍体栽培甜菜(Beta vulgar-is)A2Y的第2号染色体上。研究成功地实现了小片段低拷贝的功能基因在甜菜染色体上的定位,为今后甜菜遗传物理图谱的构建提供了有效可行的方法。
Using the cDNA fragment of MADS-box gene of sugar beet as probe, we confirmed its low-copy sequence in the genome of sugar beet by Southern blotting and initially localized it in Beta vulgar- is A2Y on chromosome 2. The successful localization of small-copy low-copy gene in beet chromosomes provided an effective and feasible method for the construction of genetic map of beet in the future.
