目的优化獐牙菜新素[(5Z)-5-乙叉基-8-羟基-3,4,5,6,7,8-六氢-1H-吡喃[3,4-c]嘧啶-1-酮,EHPO]的制备工艺,考察其保肝作用。方法经生物发酵,并采用Al_2O_3柱色谱法等色谱技术纯化、制备EHPO;采用CCl_4致小鼠急性肝损伤模型考察EHPO保肝作用。结果所建立的制备方法可获得质量分数大于95%的EHPO,药效试验表明该化合物能显著性降低小鼠血清ALT、AST水平,减少MDA量,增强SOD活力(P<0.05)结论化合物EHPO具有显著的保肝作用。
OBJECTIVE To optimize swertiacin [(5Z) -5-Ethylidene-8-hydroxy-3,4,5,6,7,8-hexahydro-1H-pyrano [3,4-c] 1-ketone, EHPO] preparation process, investigate its liver function. Methods After being fermented and purified by Al 2 O 3 column chromatography, EHPO was prepared. The model of acute liver injury induced by CCl 4 was used to investigate the protective effects of EHPO on liver. Results EHPO with mass fraction> 95% can be obtained by the preparation method. The efficacy test shows that the compound can significantly reduce the level of serum ALT and AST, decrease the amount of MDA and increase the activity of SOD in mice (P <0.05). Conclusion The compound EHPO has Significant hepatoprotective effect.
