目的探讨葛根素对肝损伤后肝细胞再生障碍大鼠外周血中T淋巴细胞亚群水平的影响。方法将40只Wistar大鼠随机分为正常组、模型组、葛根素组及护肝片组,除正常组外,其余大鼠均用2-乙酰氨基芴(2-AAF)灌胃7 d后,行肝部分切除术。术后葛根素组给予葛根素40 mg/kg灌胃给药,护肝片组给予900 mg/kg灌胃给药,正常组及模型组则给予等容积的生理盐水灌胃,连续给药7 d后,采用流式细胞术检测大鼠外周血中CD3~+、CD4~+、CD8~+、CD4~+CD25~+T细胞表达水平并计算CD4/CD8 T淋巴细胞的比值。结果与正常组比较,模型组大鼠外周血CD3~+细胞水平明显降低、CD8~+、CD4~+T淋巴细胞水平、CD4/CD8比值显著升高(P<0.05),CD4~+CD25~+T淋巴细胞水平则明显下降(P<0.01);与模型组比较,葛根素治疗组CD3~+、CD8~+、CD4~+CD25~+T淋巴细胞水平显著升高(P<0.05,P<0.01),CD4/CD8比值显著降低(P<0.05),CD4~+T淋巴细胞水平无显著差异(P>0.05)。结论葛根素可调节损伤后肝细胞再生障碍大鼠外周血T淋巴细胞亚群平衡,促进CD4~+CD25~+T淋巴细胞的升高,从而维持免疫耐受保护肝脏。
Objective To investigate the effect of puerarin on the level of T lymphocyte subsets in peripheral blood of rats with hepatocellular aplasia after hepatic injury. Methods Forty Wistar rats were randomly divided into normal group, model group, puerarin group and Hugan tablet group. All the rats except the normal group were intragastrically administered with 2-AAF for 7 days , Partial hepatectomy. Puerarin was given intragastrically in the group of puerarin 40 mg / kg orally, gavage in the liver tablet group was administered orally in 900 mg / kg, normal group and model group were given normal saline by intragastric administration, d, the expression of CD3 ~ +, CD4 ~ +, CD8 ~ +, CD4 ~ + CD25 ~ + T cells in peripheral blood of rats were detected by flow cytometry and the ratio of CD4 / CD8 T lymphocytes was calculated. Results Compared with the normal group, the levels of CD3 ~ + in peripheral blood of model group were significantly decreased, the levels of CD8 ~ +, CD4 ~ + T lymphocytes and CD4 / CD8 were significantly increased (P <0.05) (P <0.01). Compared with the model group, the levels of CD3 +, CD8 + and CD4 + CD25 + T lymphocytes in the Puerarin-treated group were significantly increased (P <0.05, P <0.01). The ratio of CD4 / CD8 was significantly lower (P <0.05) and the level of CD4 ~ + T lymphocytes was not significantly different (P> 0.05). Conclusion Puerarin can regulate the balance of T lymphocyte subsets in peripheral blood of rats with hepatocellular aplasia and promote the increase of CD4 ~ + CD25 ~ + T lymphocytes in order to maintain the immune tolerance and protect the liver.
