目的探讨胃癌及癌旁组织中活化的蛋白激酶C受体-1(RACK1)、类固醇受体辅助活化因子(Src)和B淋巴细胞瘤-2(Bcl-2)基因表达的变化。方法收集华北理工大学附属医院2011年8月1日至2014年2月1日手术切除胃癌及癌旁组织标本各80份,采用免疫组织化学法及蛋白质印迹法(Western blotting)检测RACK1、Src及Bcl-2蛋白在胃癌及癌旁组织中的表达情况,分析其相关性并结合临床病理学资料进行统计学分析。结果免疫组织化学法与Western blotting检测显示,胃癌组织中RACK1表达阳性率及表达水平均低于癌旁组织,Src与Bcl-2表达阳性率及表达水平均高于癌旁组织,差异均有统计学意义(P<0.05)。在胃癌组织中,RACK1表达分别与Src、Bcl-2表达呈负相关(相关系数r值分别为-0.632、-0.754,P<0.01),Src与Bcl-2蛋白表达无相关性(r=0.217,P>0.05)。结论胃癌组织中RACK1表达明显降低,而Src与Bcl-2表达升高。
Objective To investigate the expression of activated protein kinase C receptor-1 (RACK1), steroid receptor-associated activating factor (Src) and B-lymphoblast-2 (Bcl-2) in gastric cancer and its adjacent tissues. Methods 80 cases of gastric cancer tissues and adjacent normal tissues were collected from Affiliated Hospital of North China University of Technology from August 1, 2011 to February 1, 2014. Immunohistochemistry and Western blotting were used to detect RACK1, Src and Bcl-2 protein expression in gastric cancer and adjacent tissues, analyze the correlation and clinical pathology data for statistical analysis. Results Immunohistochemistry and Western blotting showed that the positive rate and the expression level of RACK1 in gastric cancer tissues were lower than those in paracancerous tissues. The positive rate and expression level of Src and Bcl-2 in gastric cancer tissues were higher than those in paracancerous tissues Significance (P <0.05). In gastric cancer, the expression of RACK1 was negatively correlated with the expression of Src and Bcl-2 (r = -0.632, -0.754, P <0.01, respectively), but no correlation was found between Src and Bcl-2 , P> 0.05). Conclusion The expression of RACK1 in gastric cancer tissues was significantly decreased, while the expression of Src and Bcl-2 was increased.
