野生型p53基因在细胞增殖与分化的调控过程中起着重要作用。已有研究表明,人喉癌中存在p53基因的结构异常或表达异常。本实验根据这一特点,设计出能整合在人喉癌细胞染色体上并适宜表达的野生型p53基因反转录病毒重组体Pc53N2A。通过体外实验证明,这一重组体能够以基因替代的方式,恢复p53基因的功能,抑制人喉癌细胞的生长。
The wild-type p53 gene plays an important role in the regulation of cell proliferation and differentiation. Studies have shown that there is abnormal or aberrant p53 gene expression in human laryngeal cancer. According to this feature, we designed Pc53N2A recombinant wild-type p53 gene retroviral vector that can be integrated on human laryngeal cancer cell chromosome. In vitro experiments show that this recombinant can restore the function of p53 gene and inhibit the growth of human laryngeal carcinoma cells by means of gene replacement.
