目的:研究双参通冠方对急性心肌梗死模型大鼠血流动力学的影响。方法:SD大鼠分为假手术组、模型组及双参通冠低(2.5 g/kg)、中(5 g/kg)、高(7.5 g/kg)剂量组,结扎冠状动脉左前降支建立大鼠心肌梗死模型,于十二指肠给药后0、15、30、60、90、120、180 min,采用PowerLab血流动力学信号采集与分析系统,分别检测大鼠的HR、MAP、LVSP、LVEDP、LVdp/dt max、LVdp/dt min。结果:与假手术组比较,模型组各时间点HR、MAP均升高,LVSP、LVEDP、LVdp/dt max、LVdp/dt min降低(P<0.05)。与模型组比较双参组用药15 min后HR、MAP均降低,LVSP、LVEDP、LVdp/dt max、LVdp/dt min均显著升高(P<0.05)。双参通冠高剂量组LVSP、LVEDP、LVdp/dt max、LVdp/dt min改变与中低剂量组有显著差异(P<0.05)。结论:双参通冠方能增加急性心梗模型大鼠心脏搏出量、降低心肌耗氧量和血管外周阻力,从而改善心功能。
Objective: To study the effect of Shuangshen Tong Guan Fang on hemodynamics in rats with acute myocardial infarction. Methods: Sprague-Dawley rats were randomly divided into sham-operation group, model group and the two groups, 2.5 g / kg, 5 g / kg and 7.5 g / The model of myocardial infarction in rats was established. At 0, 15, 30, 60, 90, 120 and 180 minutes after duodenal administration, the PowerLab hemodynamic signal acquisition and analysis system was used to detect the HR, MAP , LVSP, LVEDP, LVdp / dt max, LVdp / dt min. Results: Compared with the sham-operation group, the HR and MAP were increased at each time point in the model group, while LVSP, LVEDP, LVdp / dt max and LVdp / dt min were decreased (P <0.05). Compared with the model group, the HR and MAP were decreased and the LVSP, LVEDP, LVdp / dt max and LVdp / dt min were significantly increased after the two-parameter treatment for 15 min (P <0.05). The changes of LVSP, LVEDP, LVdp / dt max and LVdp / dt min in Shuangshen Tongguan high dose group were significantly different from those in low dose group (P <0.05). Conclusion: Shuangshentongguan decoction can improve cardiac function in rats with acute myocardial infarction and reduce myocardial oxygen consumption and peripheral vascular resistance.
