目的:观察更年汤对肾阳虚型围绝经期模型大鼠性激素水平的影响及安全性。方法:筛选出自然围绝经期SD大鼠80只,造模为肾阳虚型,成功后随机分为更年汤组、教材方组、替勃龙组和模型组,每组各20只;另选青年及老年雌性大鼠各20只作为对照组。用药干预15天后取血,采用酶联免疫法检测大鼠血清性激素P、FSH、LH、E2、GnRH的表达并进行比较。结果:(1)与模型组比较,更年汤组、替勃龙组和教材方组均能提高大鼠血清E2、P水平(P<0.01),下调FSH水平(P<0.05),三组之间无统计学差异(P>0.05)。(2)更年汤组、替勃龙组与模型组比较可显著降低大鼠血清LH、GnRH水平(P<0.01),教材方组与模型组比较亦有下调作用(P<0.05),但其下调作用不及更年汤组、替勃龙组(P<0.05),更年汤组与替勃龙组比较差异无统计学意义(P>0.05)。结论:更年汤能提高模型大鼠血清E2、P含量,降低LH、FSH、GnRH水平。
Objective: To observe the effect of Gengnian Decoction on sex hormones in rats with perimenopausal model of deficiency of yin-yang and its safety. Methods: Eighty SD rats were screened out to be Shen Yang deficiency type. After the success, they were randomly divided into three groups: the treatment group, the teaching materials group, the tibolone group and the model group, with 20 rats in each group. Another 20 young and aged female rats were used as control group. Blood samples were taken after intervention for 15 days, and the levels of serum sex hormones P, FSH, LH, E2 and GnRH were detected by enzyme-linked immunosorbent assay and compared. Results: (1) Compared with the model group, the Gengnian decoction group, the Tibolone group and the teaching materials group all improved the level of serum E2 and P (P <0.01) and the level of FSH (P <0.05) No significant difference between (P> 0.05). (2) The serum levels of LH and GnRH in Gengnian decoction group and Tibolone group were significantly lower than those in model group (P <0.01), while those in Guben decoction group and model group were also decreased (P <0.05) The down-regulation effect was not as good as that of Gengnian decoction group and Tibolone group (P <0.05), but there was no significant difference between Gengnian decoction group and Tibolone group (P> 0.05). Conclusion: Gengnian decoction can increase the content of E2 and P in serum, decrease the levels of LH, FSH and GnRH in model rats.
