[目的]利用血型抗原免疫及瘤内注射来治疗肿瘤,通过测定激活的脾淋巴细胞对小鼠肉瘤S180细胞的体外杀伤活性及脾指数了解小鼠的免疫功能状态。[方法]将A型血型抗原腹腔注射全身免疫小鼠4次,接种S180细胞于小鼠背部,成瘤后,于瘤内注射A型血型抗原5次。并设A型血型抗原仅全身免疫组、A型血型抗原仅瘤内注射组和空白对照组。处死小鼠,计算脾指数,用MTT比色法测定脾淋巴细胞对S180细胞的体外杀伤活性。[结果]A型血型抗原全身免疫及瘤内注射组、A型血型抗原仅全身免疫组和A型血型抗原仅瘤内注射组的脾指数均高于空白对照组(P值均<0.01)。在效靶比20∶1时,A型血型抗原全身免疫及瘤内注射组小鼠脾淋巴细胞对S180细胞的杀伤率为76.27%±4.07%,显著高于A型血型抗原仅全身免疫组、A型血型抗原仅瘤内注射组和空白对照组(杀伤率分别为10.55%±2.29%,40.92%±4.98%和9.85%±2.76%),P值均<0.01。[结论]血型抗原免疫及瘤内注射后小鼠的整体免疫功能有所提高,激活的脾淋巴细胞对靶细胞有很强的杀伤活性。
[Objective] To treat the tumor by immunization and intratumoral injection of blood group antigen, and to study the immunological function of mouse sarcoma S180 cells in vitro and spleen index by measuring the activated spleen lymphocytes. [Method] Type A blood group antigens were injected intraperitoneally to mice immunized 4 times in vivo. S180 cells were inoculated on the back of the mice. After tumor formation, type A blood group antigens were injected in the tumor five times. A type of blood group antigen and only systemic immune group, type A blood group antigen only intratumoral injection group and blank control group. The mice were sacrificed and the spleen index was calculated. The killing activity of splenic lymphocytes on S180 cells was determined by MTT assay. [Results] The spleen index of systemic immunization of type A blood group and intratumoral injection group, type A blood group antigen only in systemic immunity group and type A blood group antigen only in intratumoral injection group were higher than those in blank control group (all P <0.01). When the ratio of target to target is 20: 1, the killing rate of splenic lymphocytes to S180 cells of type A blood group antigens and the intratumoral injection group is 76.27% ± 4.07%, which is significantly higher than that of type A blood group antigen only, The type A blood group antigens only injected intratumoral injection group and blank control group (killing rate were 10.55% ± 2.29%, 40.92% ± 4.98% and 9.85% ± 2.76%, P <0.01 respectively). [Conclusion] The overall immune function of mice after immunization with blood group antigen and intratumoral injection is improved. The activated spleen lymphocytes have strong killing activity on target cells.
