目的评估单纯疱疹病毒腺苷激酶(herpes simplex virus-thymidine kinase,HSV-tk)基因对髓母细胞瘤细胞系Daoy的杀伤作用。方法 HSV-tk基因稳定转染人的髓母细胞瘤细胞系Daoy,获得Daoy-tk细胞;Daoy-tk细胞与Daoy细胞按不同比例混合,给予不同浓度的更昔洛韦(ganciclovir,GCV),7天后应用噻唑蓝(MTT)法测定细胞生存率。结果 Daoy-tk在GCV浓度为3μg/ml时,即可对Daoy细胞产生最大杀伤作用;Daoy-tk与Daoy细胞比例达到1:4时Daoy-tk仍有明显的抗肿瘤作用。结论 HSV-tk/GCV系统对人的髓母细胞有明显的杀伤作用。
Objective To evaluate the killing effect of herpes simplex virus-thymidine kinase (HSV-tk) gene on Daoy cell line of medulloblastoma. Methods Daoy-tk cells were stably transfected with HSV-tk gene in human medulloblastoma cell line Daoy-tk. Daoy-tk cells were mixed with Daoy cells in different ratios and ganciclovir (GCV) After 7 days, the cell viability was determined by MTT assay. Results Daoy-tk could kill Daoy cells maximally when GCV concentration was 3μg / ml, Daoy-tk still had obvious anti-tumor effect when Daoy-tk and Daoy cells ratio reached 1: 4. Conclusion The HSV-tk / GCV system has a significant killing effect on human medulloblast.
