目的:分析广西巴马有长寿家族史人群外周血沉默信号调节蛋白6(SIRT6)基因的mRNA表达水平,探讨其与SIRT6基因多态性及其蛋白的关系。方法:选取广西巴马地区有长寿家族史者(长寿家族史组)137例(年龄30~106岁,男70例,女67例,汉族占6.57%,壮族和瑶族占93.43%),无长寿家族史者(无长寿家族史组)91例(年龄22~89岁,男51例,女40例,均为壮族和瑶族)。采用实时荧光定量RT-PCR方法检测两组人群外周血SIRT6基因mRNA的表达水平。结果:长寿家族史组总体和女性人群外周血SIRT6 mRNA表达水平均高于无长寿家族史组(P<0.05)。长寿家族史组SIRT6基因rs350846位点GG、CG基因型及G等位基因携带者(GG、CG)的SIRT6 mRNA水平均高于无长寿家族史组(P<0.05)。长寿家族史组SIRT6 mRNA表达水平与血清中SIRT6蛋白无相关关系(P>0.05)。结论:SIRT6基因mRNA表达水平与广西巴马人群家庭聚集性长寿相关,且G等位基因携带者的SIRT6 mRNA高表达更有利于长寿。
OBJECTIVE: To analyze the mRNA expression of SIRT6 gene in peripheral blood of people with longevity family history in Bama, Guangxi, and to explore its relationship with SIRT6 gene polymorphism and its protein. Methods: A total of 137 patients with longevity family history (longevity family history group) were enrolled in the Bama region of Guangxi. The age ranged from 30 to 106 years. There were 70 males and 67 females, accounting for 6.57% of the Han population and 93.43% of the Zhuang and Yao ethnicity. Family history (no longevity family history group) 91 cases (aged 22 to 89 years, 51 males and 40 females, both Zhuang and Yao). Real-time fluorescent quantitative RT-PCR was used to detect the expression of SIRT6 mRNA in peripheral blood of two groups. Results: The expression levels of SIRT6 mRNA in peripheral blood of the overall and female population in the longevity family history group were higher than those without the longevity family history group (P <0.05). The levels of SIRT6 mRNA in GG, CG genotypes and G allele carriers (GG, CG) at rs350846 locus of longevity family history group were higher than those without longevity family history group (P <0.05). There was no correlation between SIRT6 mRNA expression in serum and serum SIRT6 protein in the longevity family history group (P> 0.05). Conclusion: The mRNA expression of SIRT6 is associated with familial aggregation and longevity of Bama population in Guangxi. And high expression of SIRT6 mRNA in G allele is more beneficial to longevity.
