目的研究MG132对小鼠急性胰腺炎(AP)相关性肺损伤的影响。方法45只BALB/c小鼠随机分为对照组、AP组和MG132组,每组又分为3、6、12 h三个亚组,每组5只。AP组和MG132组采用雨蛙肽100μg/kg腹腔内注射,1次/h,共6次;对照组注射相同剂量的生理盐水。MG132组在AP诱导前30 min腹腔注射MG132 10 mg/kg。检测血淀粉酶,免疫组化法分析肺组织核因子-κB(NFκ-B)的表达,观察肺组织病理学改变。结果与AP组比较,MG132各亚组血清淀粉酶水平均明显下降,肺组织NFκ-B表达明显下调,肺组织病理评分明显降低。结论MG132对AP相关性肺损伤有保护作用,可能与抑制NF-κB的活化有关。
Objective To investigate the effect of MG132 on lung injury in mice with acute pancreatitis (AP). Methods 45 BALB / c mice were randomly divided into control group, AP group and MG132 group. Each group was divided into three subgroups: 3, 6 and 12 h, with 5 rats in each group. The rats in AP group and MG132 group were injected intraperitoneally with Hyridase 100 μg / kg intraperitoneally once a day for 6 times. The control group received the same dose of saline. MG132 group was intraperitoneally injected with MG132 10 mg / kg 30 min before AP induction. Blood amylase was detected. The expression of nuclear factor-κB (NFκ-B) in lung tissue was analyzed by immunohistochemistry. Pathological changes of lung tissue were observed. Results Compared with the AP group, serum amylase levels were significantly decreased in all subgroups of MG132, the expression of NFκ-B in lung tissue was significantly decreased, and the pathological score of lung tissue was significantly decreased. Conclusion MG132 has a protective effect on AP-related lung injury, which may be related to the inhibition of NF-κB activation.
