利用多聚酶链式反应的新技术,建立了一种快速高效而且经济的鉴定植物基因型的方法。随意启动多聚链式反应(AP—PCR)不需DNA序列信息就可以从任一器官产生能复制的遗传指纹。这些指纹主要包括DNA片段多态性,能用于:(1)品种鉴定和亲缘测定;(2)跟踪杂交分离群体;(3)用作遗传图谱构建的标记;(4)用于产生亲缘关系(尤其在种内水平上)的差异标记。AP—PCR只需很微量的DNA(每次反应10—25ng),所以能用于DNA量有限的情况。在双未知样晶试验里,我们证明了Ap—PCR对鉴定杂交玉米植株自交系亲本的作用。
Using the new technology of polymerase chain reaction, a rapid, efficient and economical method of identifying plant genotypes was established. Feel free to initiate multi-chained reactions (AP-PCR) Generate reproducible genetic fingerprints from any organism without DNA sequence information. These fingerprints mainly include DNA fragment polymorphisms, which can be used for: (1) variety identification and kinship determination; (2) tracking hybridization segregation populations; (3) markers for genetic map construction; (4) (Especially at the intraspecific level). AP-PCR requires only a very small amount of DNA (10-25 ng per reaction) and can therefore be used in situations where the amount of DNA is limited. In the double unknown sample experiment, we demonstrated the effect of Ap-PCR on the identification of parents of inbred lines of crossed maize plants.
