目的探讨肿瘤抑素相关肽T42对人脐静脉内皮细胞和人胃腺癌细胞的抑制作用。方法本次研究开展的时间段为2016年1月~2016年6月,在大肠杆菌中表达肿瘤抑素相关T42肽,同时采用几丁质亲和层析纯化。研究所采用的方法为四甲基偶氮唑盐比色法,研究的对象为10~200mg/L肿瘤抑素相关肽(包括T42,T21,T19,T19+T21半量),与顺铂进行联合,对人脐静脉内皮细胞,以及人SGC-7901胃腺癌细胞生长所产生的影响。对40mg/L的T42肽的作用时间达到24h后,促使细胞形态所发生的变化采取苏木精-伊红染色进行检测,同时开展划痕修复实验,对40mg/L的T42肽,以及40mg/L的T19+T21肽进行半量联合使用,时间为24h,以及48h后,对细胞迁移所产生的作用。结果 T42肽不仅会抑制人脐静脉内皮细胞,同时也会抑制人SGC-7901胃腺癌细胞,同时这种抑制作用与剂量存在明显的关系;T42肽会导致两种细胞和出现凋亡现象,同时同周围的细胞发生分离,出现了皱缩,细胞的形状呈现为卵圆形或圆形;T42肽对人脐静脉内皮细胞和人胃腺癌细胞的迁移产生明显的抑制作用。同时这种抑制作用同40mg/L的T19+T21肽半量联合使用差异并不明显(P>0.05)。结论肿瘤抑素相关肽T42对人脐静脉内皮细胞和人胃腺癌细胞的抑制作用显著。
Objective To investigate the inhibitory effect of tumor suppressor-related peptide T42 on human umbilical vein endothelial cells and human gastric adenocarcinoma cells. Methods The time span of this study was from January 2016 to June 2016. The t42T42 peptide was expressed in E. coli and purified by chitin affinity chromatography. The method used in this study was tetramethylzirconate colorimetry. Results T42 peptide not only inhibited human umbilical vein endothelial cells but also inhibited human gastric adenocarcinoma SGC-7901 cells. At the same time, the inhibitory effect was not obvious with the combined use of 40 mg / L T19 + T21 peptide in half amount (P> 0.05). Conclusion Tumor suppressor-related peptide T42 has a significant inhibitory effect on human umbilical vein endothelial cells and human gastric adenocarcinoma cells.
