目的研究熊果酸(UA)对舌鳞癌细胞增殖与凋亡的作用及其可能机制。方法人舌鳞癌细胞Tca8113分别加入UA 0、10、20、40μmol/L,培养12-72h,采用MTT法检测细胞增殖,Hoechst 33342染色观察细胞凋亡的形态学变化,Western blot法检测药物作用后细胞中环氧化酶2(COX-2)、B细胞淋巴瘤-白血病2(Bcl-2)、Bcl相关X蛋白(Bax)及半胱天冬氨酸蛋白酶3(Caspase-3)的蛋白表达。结果 UA呈剂量依赖性地抑制舌鳞癌细胞Tca8113增殖,诱导舌鳞癌细胞凋亡,下调Tca8113细胞中COX-2蛋白表达,上调Bax/Bcl-2比值及活化的Caspase-3蛋白表达。结论 UA可能通过下调COX-2的表达抑制舌鳞癌细胞增殖,诱导凋亡。
Objective To study the effect of ursolic acid (UA) on proliferation and apoptosis of tongue squamous cell carcinoma and its possible mechanism. Methods Human tongue squamous cell carcinoma cell line Tca8113 was added with UA at 0, 10, 20 and 40μmol / L respectively for 12-72h. Cell proliferation was detected by MTT assay. Morphological changes of apoptosis were observed by Hoechst 33342 staining. The expression of COX-2, Bcl-2, Bcl-2 and Caspase-3 were detected by Western blot. Results UA dose-dependently inhibited the proliferation of tongue squamous carcinoma cell line Tca8113, induced the apoptosis of tongue squamous cell carcinoma, decreased the expression of COX-2 protein and up-regulated the ratio of Bax / Bcl-2 and activated Caspase-3 protein in Tca8113 cells. Conclusion UA may inhibit the proliferation and induce apoptosis of tongue squamous cell carcinoma by down-regulating the expression of COX-2.
