采用光电换能记录方法观察了槐定碱对体外培养乳鼠心肌细胞搏动的影响。槐定碱5μM时,使心肌细胞搏动频率减慢19.4±11%,搏动振幅加大24±12%,预先给予10μM哌唑嗪,可拮抗其负性频率与正性变力作用。预先给予心得安1.0μM则不影响其正性变力作用,但增强其负性频率作用。槐定碱10μM时,搏动频率加快164±112%,振幅加大26.2±9.8%;15μM时,搏动频率与振幅均减弱而停搏.提示槐定碱的作用与激动心肌细胞的α和β受体有关。
The effect of sophoridine on the beating of cardiomyocytes in neonatal rat was observed by photoelectric transduction recording method. When 5μM of sophoridine was used, the beating frequency of cardiomyocytes was reduced by 19.4±11%, and the beating amplitude was increased by 24±12%. Preemptive administration of 10μM prazosin could antagonize its negative frequency and positive inotropic effect. Pre-administration of propranolol 1.0 μM did not affect its positive inotropic effect, but enhanced its negative frequency effect. At 10 μM of sophoridine, the beating frequency was increased by 164 ± 112% and the amplitude was increased by 26.2 ± 9.8%. At 15 μM, the beating frequency and amplitude both weakened and was arrested. It is suggested that the effect of sophoridine and the α and β of cardiac myocytes Body related.
