用咖啡因作代谢探针测定研究了203例健康志愿者的N-乙酰化表型。根据咖啡因代谢物5-乙酰氨基-6-甲酰氨基-3-甲基尿嘧啶(AFMU)和甲黄嘌呤(1X)的峰高比值计算确定人乙酰化代谢表型。AFMU和1X峰高比值概率分布直方图呈明显双峰分布,区分快慢N-乙酰化表型的临界点为1.10。概率单位图与直方图结论一致。快、慢N-乙酰化表型个体分别为149例(73.4%)和54例(26.6%)。证实国人乙酰化表型分布呈多态性,与异烟肼、氨苯砜和磺胺二甲嘧啶法所测结果一致。
N-acetylated phenotypes were studied in 203 healthy volunteers using caffeine as a metabolic probe. The human acetylated metabolic phenotype was determined on the basis of the peak height ratio of the caffeine metabolites, 5-acetamido-6-carboxamido-3-methyluracil (AFMU) and mexanthine (IX) The bimodal distribution of probability distribution histograms of AFMU and 1X peak-to-peak ratio showed a bimodal distribution, and the critical point for differentiating the N-acetylated phenotype was 1.10. The probability unit chart is consistent with the histogram. The fast and slow N-acetylated phenotypes were 149 (73.4%) and 54 (26.6%), respectively. Confirmed that the national distribution of acetylated phenotypes were polymorphic, and isoniazid, dapsone and sulfamethazine test results agreed.
