目的血管内皮生长因子(VEGF)可以导致强烈的血管渗漏。越来越多的文献报道在糖尿病视网膜病变时VEGF升高。早期糖尿病性视网膜病变的一个特点是血管渗漏增加。本文的目的是探讨应用可溶性Flt阻断VEGF的作用能否抑制视网膜血管渗漏。方法重约200g的S-D大鼠通过注射链左霉素制备糖尿病模型。1周后通过静脉注射5%的PBS加甘油,或白介素-6受体单克隆抗体,或可溶性Flt,24h后应用伊凡思蓝法测定血管渗漏。用枸橼酸钠缓冲液处理的大鼠作为对照组。结果糖尿病大鼠的血管渗漏较对照组显著升高(P<0.05)。PBS加甘油和白介素-6受体单克隆抗体处理后的糖尿病大鼠的视网膜血管渗漏无变化(P>0.05),但用可溶性Flt处理糖尿病大鼠的血管渗漏显著降低(P<0.0001)。结论VEGF在糖尿病性视网膜病变的发生过程中起着十分重要的作用。通过抑制VEGF可以降低早期糖尿病性视网膜病变的血管渗漏。
Purpose Vascular endothelial growth factor (VEGF) can cause intense vascular leakage. A growing body of literature reports elevated VEGF in diabetic retinopathy. A characteristic of early diabetic retinopathy is increased vascular leakage. The purpose of this article is to investigate whether the use of soluble Flt to block VEGF suppresses retinal vascular leakage. Method S-D rats weighing approximately 200 g were prepared by injection of streptozotocin in a diabetic model. After 1 week, 5% PBS plus glycerol, or interleukin-6 receptor monoclonal antibody, or soluble Flt was injected intravenously at 5%, and vascular leakage was measured by Evans blue method 24h later. Rats treated with sodium citrate buffer served as controls. Results The vascular leakage of diabetic rats was significantly higher than that of the control group (P <0.05). There were no changes in retinal vascular leakage in diabetic rats treated with PBS plus glycerol and interleukin-6 receptor monoclonal antibodies (P> 0.05), but vascular leakage was significantly decreased in diabetic rats treated with soluble Flt (P <0.0001) . Conclusion VEGF plays a very important role in the pathogenesis of diabetic retinopathy. Vascular leakage of early diabetic retinopathy can be reduced by inhibiting VEGF.
