目的 探讨转化生长因子 (TGF β1)和口腔鳞癌侵袭转移的关系。方法 采用3 H TdR掺入法和免疫组化法观察不同浓度的TGF β1作用于口腔鳞癌TSCCa细胞系及颈淋巴转移癌GNM细胞系后细胞增殖能力和C myc蛋白表达的变化。结果 相同浓度的TGF β1对两细胞系作用不同 ;TGF β1可促进GNM细胞3 H TdR掺入率 ,同时GNM细胞中C myc蛋白表达明显上调 ;而TGF β1可抑制TSCCa细胞的增殖 ,TGF β1作用于TSCCa细胞后C myc蛋白表达水平开始下降 ,并与TGF β1有剂量依赖关系。结论 TGF β1可能在不同转化的口腔鳞癌细胞中的作用不同
Objective To investigate the relationship between transforming growth factor β1 (TGF β1) and invasion and metastasis of oral squamous cell carcinoma. Methods 3 H TdR incorporation and immunohistochemistry were used to observe the changes of cell proliferation and expression of C-myc in different concentrations of TGF-β1 in oral squamous cell carcinoma TSCCa cell line and cervical lymph node metastasis cell line GNM. Results The same concentration of TGFβ1 had different effect on the two cell lines; TGFβ1 promoted the incorporation rate of 3 H TdR in GNM cells and significantly increased the expression of C-myc in GNM cells; TGFβ1 inhibited the proliferation of TSCCa cells and TGFβ1 The expression of C-myc protein in TSCCa cells began to decline, and a dose-dependent manner with TGF β1. Conclusion TGF β1 may play different roles in different transformed oral squamous cell carcinoma cells
