目的:研究主要理化因子对西洋参细胞悬浮培养的影响。方法:利用组织培养技术结合高效液相色谱法和紫外分光光度法,考察接种量、培养基种类、基质pH和光照条件对西洋参悬浮细胞生长,人参皂苷Re,Rb1以及西洋参多糖含量的影响。结果:当接种量为25g·L-1时,西洋参细胞的干重增殖倍数显著增加;通过考察MS,SH,B53种培养基对西洋参细胞的影响,结果表明MS培养基最有利于西洋参细胞生长,B5培养基最有利于人参皂苷和西洋参多糖的合成。3种培养基中西洋参细胞多糖含量均高于栽培西洋参;pH变化对西洋参细胞生长影响不大,pH6.0时,最有利于人参皂苷Re和西洋参多糖的合成;光照培养显著促进西洋参细胞次生代谢物的合成,但对多糖合成没有太大影响。结论:接种量、培养基种类、基质pH和光照条件对西洋参悬浮细胞生长,人参皂苷Re,Rb1以及西洋参多糖合成有显著影响。
Objective: To study the effects of major physico-chemical factors on suspension culture of American ginseng cells. Methods: The effects of inoculum size, medium type, substrate pH and light conditions on the growth of ginseng suspension cells, the contents of ginsenoside Re, Rb1 and ginseng polysaccharides were studied by tissue culture technique combined with high performance liquid chromatography and ultraviolet spectrophotometry. Results: When the inoculum size was 25 g · L-1, the multiplication of dry weight of American ginseng cells was significantly increased. By examining the effects of MS, SH and B medium on American ginseng cells, the results showed that MS medium was the most conducive to the growth of American ginseng cells , B5 medium is most conducive to the synthesis of ginsenosides and polysaccharides of American ginseng. The content of polysaccharide in American ginseng cells was higher than that in American ginseng cultivated in three kinds of media. The change of pH had little effect on the growth of American ginseng cells. At pH 6.0, it was most conducive to the synthesis of ginsenoside Re and American ginseng polysaccharide. Metabolites synthesis, but not much effect on polysaccharide synthesis. CONCLUSION: Inoculum size, medium type, substrate pH and light conditions have significant effects on the growth of suspension cells, ginsenoside Re, Rb1 and polysaccharide synthesis of American ginseng.
