利用家栽和野生中国黄连的幼叶切块,接种在含有1.0~2.0mg/L2,4-D和0.1mg/LKT的6,7-V固体培养基上,诱导形成愈伤组织。选择较松散的愈伤组织转入液体悬浮培养,获得游离细胞和细胞聚集体。经60Co-γ射线辐射诱变和平板培养,筛选出小檗碱含量相当于6年生亲本植物根茎含量的有8个细胞株(系),最高的细胞系含量为6.12%。
The young leaves of Chinese and Coptis chinensis were cut and inoculated on 6,7-V solid medium containing 1.0-2.0 mg / L 2,4-D and 0.1 mg / L KT to induce the formation of callus organization. Choose loose callus into liquid suspension culture, to obtain free cells and cell aggregates. After 60Co-γ-ray irradiation and plate culture, eight cell lines (lines) with the berberine content equivalent to that of the 6-year-old parent plants were screened, and the highest cell line content was 6.12%.
