目的探讨体外腺病毒介导的白细胞介素-24基因(Ad-IL-24)对间变性大细胞淋巴瘤(ALCL)Karpas299细胞增殖及凋亡的影响。方法将Karpas299细胞分为空白对照组、Ad-IL-24组及携带绿色荧光蛋白基因的重组腺病毒组(Ad-GFP组)。Ad-IL-24组加入200.0μL Ad-IL-24原液,Ad-GFP组加入200.0μL Ad-GFP原液,空白对照组加入200.0μL PBS溶液。分别于处理12、24及48 h后,采用细胞毒性与增殖检测试剂盒(CCK-8试剂盒)检测3组的细胞增殖抑制率;于处理48 h后,采用流式细胞仪检测3组的细胞凋亡率。结果 Ad-IL-24对Karpas299细胞的生长具有抑制作用,且随时间延长,Ad-IL-24组的细胞增殖抑制率呈上升趋势;同时点与Ad-GFP组相比较,12、24及48 h时Ad-IL-24组的细胞增殖抑制率均较高(P<0.05)。与空白对照组及Ad-GFP组相比较,Ad-IL-24组的细胞凋亡率较高(P<0.05)。结论 Ad-IL-24对Karpas299细胞的生长具有抑制作用,且能引起Karpas299细胞的凋亡。
Objective To investigate the effect of adenovirus-mediated interleukin-24 gene (Ad-IL-24) on proliferation and apoptosis of amelobular large cell lymphoma (ALCL) Karpas299 cells. Methods Karpas299 cells were divided into blank control group, Ad-IL-24 group and recombinant adenovirus group (Ad-GFP group) carrying green fluorescent protein gene. Ad-IL-24 group 200.0μL of Ad-IL-24 stock solution, 200.0μL Ad-GFP stock solution and Ad-GFP group 200.0μL PBS solution. After 12, 24 and 48 h treatment respectively, the cell proliferation inhibition rate of the three groups was detected by the cytotoxicity and proliferation assay kit (CCK-8 kit); After 48 h of treatment, Rate of apoptosis. Results Ad-IL-24 had an inhibitory effect on the growth of Karpas299 cells. With the prolongation of time, the cell proliferation inhibition rate of Ad-IL-24 group showed an upward trend. Compared with Ad-GFP group, 12, 24 and 48 h Ad-IL-24 group of cell proliferation inhibition rate was higher (P <0.05). Compared with the blank control group and Ad-GFP group, the apoptosis rate of Ad-IL-24 group was higher (P <0.05). Conclusion Ad-IL-24 can inhibit the growth of Karpas299 cells and induce the apoptosis of Karpas299 cells.
