目的:观察知柏地黄汤合桃红四物汤对CsA肾毒性模型大鼠的肾保护作用。方法:予30mg/kg剂量的CsA灌胃制作大鼠CsA肾毒性模型,同时分别予10mg/kg贝那普利、2g/mL中药药液大鼠灌胃以预防肾毒性。予4周末分别观察各组大鼠肾功能、肾组织病理变化,免疫组化观察各组大鼠肾组织TGFβ1的表达变化。结果:与模型组相比,中药组大鼠血肌酐降低(28.3±3.8 VS 33.1±5.2,P=0.052),病理显示中药组大鼠肾组织空泡变性较模型组改善[变性范围(8.2±2.2)%VS(15.6±3.7)%,P<0.05],炎症细胞浸润及间质纤维化亦减轻;免疫组化显示,中药组大鼠肾组织TGFβ1的表达降低。结论:知柏地黄汤合桃红四物汤具有保护和改善CsA慢性肾毒性大鼠肾功能的作用。其机制有可能与降低肾组织TGFβ1的表达有关。
OBJECTIVE: To observe the renal protective effect of Zhibo Di Huang Tang combined with Taohong Siwu Decoction on CsA nephrotoxicity model rats. Methods: The CsA nephrotoxicity model was established by intragastric administration of CsA at a dose of 30 mg / kg. At the same time, rats were intragastrically administered with 10 mg / kg benazepril and 2 g / mL Chinese medicine to prevent nephrotoxicity. At the end of 4 weeks, the renal function and renal pathological changes of rats in each group were observed. The expression of TGFβ1 in renal tissue of each group was observed by immunohistochemistry. Results: Compared with the model group, the serum creatinine of the Chinese medicine group decreased (28.3 ± 3.8 VS 33.1 ± 5.2, P = 0.052). The pathological changes showed that the degeneration of renal tissue in the TCM group was improved compared with the model group [range of degeneration 2.2% VS (15.6 ± 3.7)%, P <0.05]. Inflammatory cell infiltration and interstitial fibrosis also alleviated. Immunohistochemistry showed that the expression of TGFβ1 was decreased in the kidney of TCM group. Conclusion: Zhibai Dihuang Decoction combined with Taohongsiwutang can protect and improve renal function of rats with chronic nephrotoxic CsA. Its mechanism may be related to reducing the expression of TGFβ1 in the kidney tissue.
